Matters needing attention in operation of sterile inoculating loop

Bacteria inoculation inspection operation must strictly abide by the relevant systems and operating procedures, strengthen the concept of disinfection and sterility, so as to avoid accidents.
The following points should be paid attention to when using the inoculation ring in the specific experimental operation:
1. Disposable inoculating loops must be used when observing the morphology of bacteria from the tested specimen or culture. The inoculation ring is made of a nickel resistance wire or special platinum wire with a length of about 5 ~ 8 cm and moderate hardness, which is placed on a metal or glass rod. Those without rings are called vaccination needles.
2. Before and after each use of the inoculating loop and needle, flame sterilization is required. That is to say, burn thoroughly on the flame of ethanol lamp once, and the metal rod or glass rod part must also rotate through the flame for three times. When burning after use, make the heat guide to the inoculation ring. If the ring is burned directly, the residual bacteria liquid on the ring can be suddenly and violently splashed by high heat, which is dangerous for infection. After the bacteria liquid on the ring evaporates and dries up, the inoculation ring is burned and sterilized in the flame in the vertical direction, and finally the metal rod or glass rod is partially reciprocated through the flame. The inoculation needle is sterilized after use in the same way as the inoculation ring.
3. After the inoculating loop and needle passes through the flame, it is necessary to cool before taking samples or placing them on the workbench to prevent scalding microorganisms and burning the desktop.
