1. Number of holes
Most users choose to conduct tissue culture on culture flasks. However, many applications require growth plates. Expanding to 384-well plates requires even more automated applications.
2. The shape of the hole
The bottom of the well can be flat bottom, U bottom, V bottom, depending on the cell type and downstream applications. For traditional 2D cell culture (such as HeLa or MDCK cells), especially when imaging or spectrophotometric measurement of the culture is required, F-bottom is usually preferred. For cells without contact inhibition, C-bottom is also good. U-bottom is suitable for suspension culture (such as spheroid cell culture), because the round surface makes it difficult for cells to attach and grow. The V-type (cone bottom) culture plate is mostly used for immunological hemagglutination experiments.
3. The color of the culture plate
The color of the culture plate is also closely related to the application. If you observe the cells with a phase-contrast microscope or with the naked eye, you can choose a transparent culture plate. However, for applications outside the visible light spectrum (such as luminescence or fluorescence), colored perforated plates (such as white or black) are necessary. Cold light samples usually choose a white surface to maximize signal reflectivity, while fluorescent applications greater than 300 nm usually use a black surface to absorb the excitation signal. The colored surface can also prevent signal crosstalk between adjacent holes.
4. Surface treatment
The choice of cell surface treatment depends on whether you are culturing suspension cells or adherent cells.
